چکیده مقاله:

In this paper, a fast and sensitive localized surface plasmon resonance (LSPR) based biosensor was developed and the optimization of gold – antibody conjugates through investigation of different parameters were performed. Gold nanoparticles (AuNPs) with a size of ~20 nm were synthesized via chemical reduction of HAuCl4 with trisodium citrate as reducing and stabilizing agent. The impacts of pH of gold colloids and antibody concentrations on conjugation of electrostatically absorbed antibodies on the AuNPs surface were evaluated. The diverse amounts of antigens were added to the selected gold – antibody conjugate and the calibration curve and limit of detection of the system were successfully obtained. The UV- Vis and DLS outputs were utilized to prove the efficiency and repeatability of the system. As a result, the designed biosensor shows a convincing LOD of 400 ng/ml of antigen. It is suggested that electrostatic absorption strategy of AuNPs with negative charge to the positively charged antibodies can be an efficient methodology. Results showed an effective LSPR system for detection of small amounts of antigen in short time as well as with high accuracy.

Surface plasmon resonance and magnetism of thiol-capped gold nanoparticles.

Surface plasmon resonance measurements and magnetic characterization studies have been carried out for two types of thiol-capped gold nanoparticles (NPs) with similar diameters between 2.0 and 2.5 nm and different organic molecules linked to the sulfur atom: dodecanethiol and tiopronin. In addition, Au NPs capped with tetraoctyl ammonium bromide have also been included in the investigation sinc...

Applications of Surface Plasmon Resonance (SPR) for the Characterization of Nanoparticles Developed for Biomedical Purposes

Great interest is currently being devoted to the development of nanoparticles (NPs) for biomedical purposes, designed to improve the pharmacokinetic profile of their cargos (either imaging probes or drugs) and to enhance the specific targeting at the disease site. Recent works suggest that Surface Plasmon Resonance (SPR), widely used for the analysis of biomolecular interactions, represents a t...

A Gold Nanoparticles Enhanced Surface Plasmon Resonance Immunosensor for Highly Sensitive Detection of Ischemia-Modified Albumin

In this study a novel sensitive nanogold particle sensor enhancement based on mixed self-assembled monolayers was explored and used to construct a Surface Plasmon Resonance (SPR) immunosensor to detect Ischemia Modified Albumin (IMA). Compared with a direct binding SPR assay at a limit of detection (LOD) of 100 ng/L, gold nanoparticles (AuNPs) of 10 nm dramatically improved the LOD of IMA to 10...

Early diagnosis of oral cancer based on the surface plasmon resonance of gold nanoparticles

The high mortality rate in cancer such as oral squamous cell carcinoma is commonly attributed to the difficulties in detecting the disease at an early treatable stage. In this study, we exploited the ability of gold nanoparticles to undergo coupled surface plasmon resonance and set up strong electric fields when closely-spaced to improve the molecular contrast signal in reflectance-based imagin...
